Sedimentation velocity analytical ultracentrifugation (SV-AUC) method is the established standard experiment when it comes to AUC. Interaction Parameter Determination

In the last couple of years, the interest in the determination of predictive parameters within formulation development has grown. Interactions parameters provide valuable information for the prediction of molecular stability. These values are used in a “design of experiment” (DOE) approach to assist formulation developments. At ZentriForce Pharma we focus on the determination of molecule – molecule and molecule – formulation interactions. Determination of Molecule – Molecule Interactions

If the focus is solely on molecule – molecule interaction, the determination of the second osmotic virial coefficient (B22) via sedimentation equilibrium analytical ultracentrifugation (SE-AUC) is highly recommended. Alternatively, it is possible to determine the concentration dependence of the sedimentation coefficient (ks). Both, ks and B22 are in all cases positive as they solely describe the repulsive molecule – molecule interaction. We are able to provide high resolution and high throughput ks and B22 determinations  because we carry out all measurements on the new Optima Analytical Ultracentrifuge

Determination of Molecule – Formulation Interactions

If the aim of interaction parameter determination is to include the molecule –formulation interaction, the determination of the second osmotic virial coefficient (A22) via static light scattering (SLS) is recommended. Even though the name of this parameter is identical to B22, A22 describes additional interactions. Furthermore, the concentration dependence of the diffusion coefficient (kd) is assessable. Both, A22 and kd can be either positive or negative. They are mixtures of the repulsive molecule – molecule (always positive) and the molecule – formulation interaction (can be negative). With the DynaPro®Plate Reader III ZentriForce Pharma performs measurements on an instrument that was optimized for this purpose. We can provide both A22 and kd from a single experiment as we are able to simultaneously collect DLS and SLS data. 

Owing to a well-plate system the instrument allows high-throughput campaigns of different formulations, thus circumventing the time intensive procedure of a cuvette-based instrument. 

Minimum sample volume is 4 µl without the requirement to introduce an oil film to cover the sample.
