Particle Size Determination 

Depending on the type of your API, particle size determination may be one of the most important steps for the monitoring of your drug product’s purity. For gene therapy products especially, it is important to analyze the particle size distribution. Our laboratory offers the analytical techniques that you need for the determination of particle size distributions. Below, we provide a portfolio of techniques, including their applicable size ranges. 

Dynamic Light Scattering (1 nm – 1 μm)

With the DynaPro®Plate Reader III ZentriForce Pharma offers highest quality DLS measurements. The instrument performs dynamic light scattering (DLS) as well as static light scattering (SLS) measurements. Its properties allow for drastic reduction of sample volume, with a minimum of 4 µL, without the need to introduce an oil film to cover the sample. Additionally, sample measurement at low concentrations is feasible, owing to the instrument’s high sensitivity.

Additionally, the instrument is equipped with a well-plate system that allows high throughput campaigns of up to 1536 samples in a single run. We can therefore assess the particle size distribution of different samples simultaneously, for example within a formulation development.

Asymmetrical Flow Field-Flow Fractionation (1 nm – 1 μm)

Asymmetrical flow field-flow fractionation (AF4) is an excellent technique for the determination of particle sizes in the considerable size range of 1 nm to 1 µm. AF4 is a first principle method that determines the hydrodynamic radius.). The combination of the fractionation method itself (AF4) with the use of UV, RI and MALLS detectors allows us to get the full picture about the measured samples.

    • radius of gyration 
    • molecular weight 
    • UV signals at two wavelength

    • light scattering signal 

Analytical Ultracentrifugation (0.5 nm – 200 nm)

Analytical Ultracentrifugation (AUC) is an established technique for the determination of particle size distributions. It was originally developed for this very purpose nearly 100 years ago. Its properties are especially interesting for gene therapy products. AUC can separate species by size as well as density. This unique property gives access to information on sample purity (it detects fragments and aggregates) and on the ratio of particles that contain DNA to empty particles within a single measurement (applicable to, e.g., AAVs).
